summaryHeterozygous mutations in NADP-dependent isocitrate dehydrogenases 1 and 2 (IDH) define the large majority of diffuse gliomas and are associated with hypermethylation of DNA and chromatin. The exact oncogenic process and the metabolic aberrations imposed by mutant IDH remain however poorly understood. Studies on IDH mutant gliomas are hampered by difficulties to maintain these cells in culture. We have generated and characterized orthotopic patient-derived glioma xenografts carrying the endogenous IDH mutation, allowing to profile the distribution of metabolites in situ using high resolution mass spectrometric imaging (MSI) and liquid chromatography mass spectrometry (LC-MS) (Fack et al. EMBO Mol Med 2017). The metabolic landscape of IDH mutant gliomas with respect to phospholipid, energy and oxidative stress pathways will be discussed, in light of their potential to novel metabolic vulnerabilities in these tumors.
