summaryVitamin A5/X, a recently identified new class of vitamin A has been proposed to control episodic memory, but the exact cellular and physiological substrates of such modulations remain unknown. We have shown that 9-cis-13,14-dihydroretinoic acid (9CDHRA), an active form of vitamin A5/X is an endogenous ligand of Retinoid X Receptors (RXRα, β, γ), a group of nuclear hormone receptors acting as ligand-regulated transcription factors. We found that RXRγ is functionally predominant RXR in mediating pro-mnemonic activities of 9CDHRA in spatial working memory tasks. Using whole-cell recordings and somatic ablation of RXRγ in the hippocampus in mice we determined that RXRγ controls activities of hilar mossy cells), a group of hilar excitatory interneurons in dentate gyrus and that such regulation may be relevant for control of pattern separation and working memory performance in mice. We speculate that such regulations might be also pertinent for understanding of the vitamin A5/X signaling in control of mnemonic functions.
